Quantitative Polymerase Chain Reaction (qPCR) is a powerful molecular biology technique used to quantify the amount of a specific DNA sequence in a sample. It builds upon the principles of traditional PCR but incorporates fluorescent dyes or probes to measure DNA amplification in real time.
qPCR technology typically involves the following procedure:
- Denaturation:
- The thermocycler heats up to roughly 95 °C.
- The double-stranded DNA separates into single strands.
- Annealing:
- The temperature lowers down to 45-65 °C.
- The single-stranded primers attach to the appropriate ends of the target sequence.
- Extension:
- The temperature is raised to around 68-72°C.
- DNA polymerase synthesizes a new DNA strand using the primers as templates.
- Cycling:
- The thermal cycles are repeated multiple times (usually 20-40 cycles).
- Each cycle doubles the amount of DNA.
- Quantification:
- The qPCR instrument with high-sensitivity spectrometers records the fluorescence emitted at the end of each amplification cycle.
